Zodpovědnost za selhání vývoje klonovaných embryí byla a stále je připisována nedostatečné či chybné reprogramaci přenášeného jádra embryonální či somatické buňky, která prochází postupnými změnami při diferenciaci pro tvorbu tkání a orgánů. Za ideálních podmínek by měl přenos jádra částečně nebo již specializované buňky navrátit jádro na úroveň zygoty vzniklé po oplození. Rozvoj molekulárních metod vedl k poznání, že aktivace genů a následná diferenciace buňky je podmíněna pouze epigenetickou modifikací jaderné DNA.

Insufficient or incorrect reprogramming of the transferred nucleus of an embryonal or somatic cell, which gradually changes during the differentiation leading to development of tissues or organs, is a supposed cause of failure in development of cloned embryos. Under ideal conditions the nucleus transfer of a partly or fully specialized cell should return the nucleus to the level of a zygote after impregnation. The development of embryonal methods has led to recognition that gene activation and the consequent cell differentiation are conditioned only by epigenetic modification of the nuclear DNA.
